# Lilly is putting $6.5 billion into concrete and up to $1 billion into AI drug discovery

> Investors moved this week to mark Lilly's AI-discovery partner Isomorphic Labs at up to $50 billion, a company that has never dosed a patient. Lilly's own ledger, $6.5 billion of concrete against at most $1 billion of AI, says which bet it trusts.

## The market marked Lilly's AI partner at $40 billion this week

On October 8, [Isomorphic Labs](https://theinference.org/markets/companies/isomorphic-labs), the [Alphabet](https://theinference.org/markets/companies/alphabet) majority-owned spinout that counts [Eli Lilly](https://theinference.org/markets/companies/eli-lilly) among its pharma partners, [was reported in early talks to raise at $40 billion to $50 billion](https://theinference.org/article/isomorphic-labs-reported-40-billion-raise-prices-biohub-s-1-8-billion-cell-data-pact), up from roughly $17.5 billion in May after a $2.1 billion Series B [1][2]. Isomorphic has never dosed a patient; first-in-human trials are targeted by the end of 2026 [1][2]. Its deals with Lilly, Novartis and Johnson & Johnson carry more than $3 billion in potential milestones, payable only on development success [1]. The risk lands on anyone pricing Lilly's AI optionality at private-market rates: the $40 billion mark belongs to a private company Lilly does not own, and Lilly's own spend on the same thesis stops at $1 billion.

## Lilly answered with $6.5 billion of concrete

On September 21, Lilly [broke ground outside Houston on a $6.5 billion plant](https://www.globalconstructionreview.com/ground-broken-on-6-5bn-eli-lilly-manufacturing-facility-for-glp-1/) to make the active ingredient for Foundayo, its oral GLP-1 pill: roughly 4,000 construction jobs, more than 600 permanent ones, and the tenth US manufacturing site Lilly has announced since 2020 [3][4][5]. Texas Governor Greg Abbott called it the largest active-pharmaceutical-ingredient project in the state's history [3]. Foundayo is approved in the US, under review in more than 40 countries, and slated for a full global rollout in 2027 [3][4].

## Mounjaro's $9.9 billion a quarter underwrites the concrete

The engine paying for it is Mounjaro, the world's top-selling drug: $9.9 billion last quarter alone, nearly $23 billion across 2025, and more than a third of Lilly's total sales [6][7]. Quarterly revenue has more than doubled since late 2023 while quarterly capital spending tripled, to $2.9 billion [8]. The franchise carries its own exposure: a North Carolina man [sued Lilly this month, claiming Mounjaro and Trulicity left him with stomach paralysis](https://www.ibj.com/articles/eli-lilly-sued-over-stomach-problems-patient-ties-to-mounjaro-and-trulicity) [6].

*Chart: **Lilly's quarterly revenue has more than doubled since 2023 while capex tripled** Line chart of Eli Lilly quarterly revenue and capital spending from Q3 2023 to Q2 2026, revenue rising from $9.5B to $23.0B and capex from $1.0B to $2.9B*

## Lilly's AI budget is one line, at most $1 billion over five years

The week's AI headlines rest on a number that has not changed since January 12: [up to $1 billion over five years](https://cryptobriefing.com/nvidia-eli-lilly-1b-ai-drug-discovery-lab/), co-invested with [Nvidia](https://theinference.org/markets/companies/nvidia) in a Bay Area lab running on BioNeMo and the coming Vera Rubin chips [9]. "Up to" is a ceiling, not a check. Nvidia's win is positional: if BioNeMo and Vera Rubin become the default stack for biology models at a drugmaker of Lilly's scale, the chipmaker holds the reference design for pharma's AI labs [9]. Lilly pays for AI like an option and for factories like a certainty. Its own shelf priced softer: TRex Bio, a Lilly-backed autoimmune biotech, [raised $116.7 million at $14 a share](https://www.bloomberg.com/news/articles/2026-10-09/eli-lilly-backed-biotech-trex-raises-116-7-million-in-us-ipo), the bottom of its range [10].

*Chart: **Lilly's concrete budget dwarfs its AI budget; the market marks its AI partner higher** Bar chart comparing Lilly's $6.5B Houston plant commitment and the up-to-$1B Nvidia lab ceiling against Isomorphic Labs' $17.5B May 2026 mark and its reported $40 to $50 billion funding talks, drawn at the $40B floor*

*Lilly cash commitments and private-market valuations, $ billions. The Nvidia figure is a five-year ceiling announced January 12, 2026; the Isomorphic in-talks bar is drawn at the reported $40B floor of the $40 to $50 billion range reported October 8, 2026. [1][2][3][9]*

## Three catalysts will settle the ledger

[Foundayo's next clinical data, in diabetes and sleep apnea, lands later this year](https://theinference.org/article/lilly-sells-a-modelled-57-diabetes-risk-cut-on-foundayo-just-as-its-drug-prices-fall) [4]. Isomorphic's first-in-human trial is due by the end of 2026 [1][2]. Foundayo's global rollout follows in 2027 [3]. Until then, the ledger reads $6.5 billion of concrete and, at most, $1 billion of code.

## Takeaway

Lilly funds AI drug discovery like an option, at most $1 billion over five years alongside Nvidia, while Mounjaro's $9.9 billion quarter underwrites $6.5 billion of GLP-1 manufacturing concrete; the $40 billion AI mark belongs to Isomorphic Labs, which Lilly does not own and which has never dosed a patient.
